Conventional imaging SPR interrogate the signal from a fixed incident angle. The signal of such system during the biosensing process is a convolution of spectra angle contour and the movement of the SPR dip shift. As shown in the lower left image, the two consecutive round of identical sensing signal, is translated to two signals with variations up to 50%. This is mainly due to the spectrum contour differences coming from fabrication variation. While manufacturing condition varies from day to day, especially in common lab setting, centroid based SPR provide more consistent performance. While centroid based is trusted by many of the commercialized SPR systems, it also has drawback in losing the capacity of large scale paralleled detection.
